What would be the file that have this information? You mean where is the manager info in the person document ? You should open your LDAP directory in edit mode ( with the appropriate tool ) and check the value of that field. In Domino is in the person doc, in AD is in the user properties, ....

Which would be the process to run to update managers? Run the population wizard, the same you ran to populate the Profiles db. There's an option to mark managers, select it.
